چکیده

Effect of tetrodotoxin (TTX) on the aconitine-induced arrhythmias of the isolated cardiac tissues of the chicken embryo, the toad and the golden carp is studied by a microelectrode technique. TTX in a dose of 10-8-10-7 g/ml antagonized the aconitine-induced rapid firing in the chicken embryo ventricle, the toad atrium and the golden carp atrium. TTX was much more potent than lidocaine in exerting an antiarrhythmic effect. The mode of action of aconitine and TTX is discussed.

Mode of action of new anti-arrhythmic agents.

Propranolol, diphenylhydantoin and tetrodotoxin showed inhibitory or removal effects on fibrillation induced in isolated rabbit atrial muscle strip. Depending upon methods of induction of fibrillation, effects of these drugs were different. The mode of their action seemed to be due to following electrical natures of each drug in its effective dose. Diphenylhydantoin decreased rate of rise and a...

Inhibition of ATP-sensitive potassium channels of adult rat heart cells by antiarrhythmic drugs.

We have investigated the effect of antiarrhythmic drugs on the increased potassium conductance induced in isolated adult rat heart cells by ATP depletion. The rate of 86Rb uptake in the presence of ouabain was used as a measure of potassium conductance. Treatment of cells with rotenone plus p-trifluoromethoxyphenylhydrazone (FCCP) rapidly depleted ATP levels and strongly stimulated the rate of ...

The effects of antiarrhythmic drugs, stimulation frequency, and potassium-induced resting membrane potential changes on conduction velocity and dV/dtmax in guinea pig myocardium.

For one-dimensional propagation, a nonlinear relationship between Vmax and conduction velocity is predicted by cable theory, and, under experimental conditions, Vmax and conduction velocity may change in opposite directions. Using standard microelectrode techniques, we have measured Vmax and conduction velocity in guinea pig papillary muscles exposed to tetrodotoxin and low sodium (agents expec...

Fipronil: mechanisms of action on various organisms and future relevance for animal models studies

Because insects had developed resistance to several insecticides, today, neonicotinoids and fiproles are used to combat pests. The difference between this two classes of insecticides is that fipronil acts by inhibiting the receptors of nervous cells, while neonicotinoids perturbs the neuronal transmission. The suitable properties of fipronil make that its use to be more obvious on the pesticide...

The antiarrhythmic drug BRL-32872.

BRL-32872 is a new antiarrhythmic drug with balanced class-III and class-IV actions as categorized by the Vaughan-Williams classification. BRL-32872 blocks the rapid component of the cardiac delayed rectifier potassium channel IK(r) (IC(50) = 28 nM) and its molecular correlate HERG ("Human-ether-a-go-go related gene," IC(50) of 19.8 nM in cell lines) at low concentrations. It also inhibits the ...
